Whether the drug improves hard clinical outcomes is not settled. No completed trial has shown a reduction in heart attacks or strokes among treated patients, although a dedicated cardiovascular outcomes study has been discussed in the literature. Investigators have also examined hepatic fat in people with HIV and fatty liver disease, cognitive measures in small cohorts, and changes in bone density. Regulatory labeling emphasizes monitoring of insulin-like growth factor 1 because supraphysiologic levels raise questions about tissue growth, and the clinical significance of that signal remains an open question rather than a demonstrated harm.
Binding of tesamorelin to the growth hormone-releasing hormone receptor on anterior pituitary somatotrophs activates a Gs protein pathway, raises cyclic AMP, and triggers release of stored growth hormone into the bloodstream. Because the analogue resists dipeptidyl peptidase-4, its plasma residence time exceeds that of native GHRH, producing a larger and more sustained secretory signal. The released growth hormone then acts on the liver and peripheral tissues to raise insulin-like growth factor 1, which feeds back on the hypothalamus and pituitary. This axis explains both the intended effects on fat distribution and the biological markers used to track them.

Development work on the compound, originally designated TH9507, focused on conditions in which reduced growth hormone signaling is thought to contribute to altered body composition. The United States Food and Drug Administration approved it in 2010 for the treatment of excess visceral abdominal fat in adults with human immunodeficiency virus infection and lipodystrophy. Later research examined other populations, including adults with mild cognitive impairment, where a large trial did not meet its primary endpoints. This mixed record illustrates how a single mechanism can produce clear effects in one setting and inconclusive results in another.
Several related peptides act on the same receptor, including sermorelin, a shorter GHRH fragment, and modified analogs such as CJC-1295 and modified GRF(1-29) that are common in research settings rather than approved products. Tesamorelin differs from growth hormone itself in that it acts upstream, prompting the pituitary to release the hormone through physiological signaling rather than supplying it directly. Terminology in the literature distinguishes GHRH analogs, growth hormone secretagogues, and recombinant growth hormone, although popular discussion often blurs these categories together. Precise naming matters when comparing study results.
Research supply is often accompanied by a certificate of analysis listing chromatographic purity, mass confirmation, and storage conditions. Laboratories compare that document with an independent test when material is intended for bench work, since certificates describe a batch rather than an individual vial. Published studies usually state the source and purity of the peptide because small differences in purity can shift measured activity. Full analytical validation is rarely reported, which leaves batch-to-batch comparability an open question.
The peptide is supplied as a lyophilized powder in single-use vials and is normally kept refrigerated between two and eight degrees Celsius, protected from light. Once dissolved, the solution is handled carefully because peptide bonds and the acyl modification can degrade under warm or alkaline conditions. Vials are inspected for cracks, and the powder is checked for color and uniformity before handling. Temperature excursions during shipping are a frequent reason for quality questions.
Metabolic interest in this compound centers on fat distribution rather than on hormone levels alone. Imaging trials in adults with excess abdominal fat report reductions in visceral adipose tissue, while subcutaneous depots change comparatively little. Growth hormone and IGF-1 are presumed to carry the effect, but the separate contribution of each is not firmly established. Whether these changes persist after treatment stops, and whether they alter longer-term health outcomes, remain open questions that published work does not answer consistently.
Tesamorelin is a synthetic peptide of forty-four amino acids whose sequence reproduces human growth hormone-releasing hormone. Its distinguishing feature sits at the amino terminus, where a trans-3-hexenoyl group replaces the free amine. That acylation slows cleavage by dipeptidyl peptidase IV, an enzyme that otherwise removes the first two residues and inactivates the natural hormone quickly. The modified peptide therefore persists longer in circulation while keeping the same receptor target. It is handled as a lyophilized solid and dissolved shortly before use.
Measured responses usually involve growth hormone and insulin-like growth factor 1, known as IGF-1. Growth hormone rises in bursts and is difficult to sample reliably, while IGF-1 shifts more slowly and can be assessed from a single blood draw. Studies therefore treat IGF-1 as the more practical pharmacodynamic marker. Both are indirect, showing that the receptor was engaged rather than that the peptide reached a particular concentration. Direct exposure measurement requires an assay aimed at the molecule itself.
Published work tends to frame tesamorelin as a tool for studying the GHRH axis and as a compound with measurable effects on body composition. Reports often describe visceral adipose tissue as an endpoint, assessed by imaging rather than by inference. Analytical sections commonly describe liquid chromatography with tandem mass spectrometry to confirm identity and purity, because immunoassays may cross-react with related fragments. Where results diverge between studies, differences in assay choice, sampling timing, and population are frequent explanations offered. Whether effects persist after treatment stops remains an open question.
Tesamorelin binds the growth hormone–releasing hormone receptor on pituitary somatotroph cells. The receptor signals through the Gs protein, raising intracellular cAMP and activating protein kinase A. That cascade triggers release of stored growth hormone in pulses rather than a steady stream. Because the drug acts at the receptor that normally controls this process, its effect depends on the body's own signaling architecture rather than on a synthetic pathway. The resulting hormone profile reflects the timing of each pulse, not only its size.
The 1981 Springboks tour of New Zealand was compromised by demonstrations, and the tourists had to be kept under strict security throughout their visit. Two games had to be cancelled. The game at Hamilton in the first week of the tour saw 200 protestors rip down a chain fence, sprinkle tacks all over the pitch and then staged a sit-in on the halfway line. At the time, a crowd of 25,000 was watching them playing Waikato. Subsequent matches saw the arrival of barbed wire, and police with batons. The match against South Canterbury at Timaru was cancelled because the authorities thought that they would not be able to control the demonstrations there. The final test of the tour was buzzed by a Cessna aircraft - some in fact nicknamed it the "Crazy Biggles Test". The plane continually strafed the pitch, and dropped flour bombs, flares and leaflets. All-Black prop Gary Knight was temporarily stunned by a flour bomb. Musician Bruce Russell received two police convictions as a student leader of protests against the tour. He was at the University of Otago at the time. It was a tight game, with Allan Hewson, the New Zealand full-back kicking a long range penalty to win the game 25–22. As Rod Chester and Nev McMillan described the scene:
rough endoplasmic reticulum (RER) A type of membrane in the endoplasmic reticulum with numerous ribosomes conspicuously attached to its surface, in contrast to the "smooth" endoplasmic reticulum which lacks ribosomes. The rough ER serves as the site of protein synthesis for the majority of the cell's secreted and transmembrane proteins, as well as the site of synthesis of membrane lipids. It may be continuous with the smooth ER or exist separately.
== History == Azvudine was first found in literature in a patent filed by Chang Jun-biao of Zhengzhou University. It received its current name in 2009, when researchers at Roche independently discovered it as a Hep C RNA polymerase inhibitor in vitro. In the following years, Chinese scientists tested it in vitro for a number of targets, most importantly HBV (human and duck) and HTLV-1, two viruses with a reverse transcriptase. It was first proposed as an HIV treatment in 2011, when in vitro tests by the Chang group provided positive results. In 2014, its oral pharmokinetics in rats was elucidated. A phase II study (NCT04109183) was finished in March 2019 by Genuine Biotech. In August 2020, the Chang group found that the substance inhibits vif in vitro. In the same month, China's drug regulator (NMPA) decided to fast-track the approval process, labelling it a first-in-class medication. In July 2021, NMPA granted conditional approval for HIV-1. It was included in the 2021 HIV treatment recommendnation by the Chinese Medical Association and Chinese CDC, published October that year. Curiously, no full results of the trial have been made available for this study in any journal detailing the experiment design as of December 2022. Parts of the results are shown on the drug monograph as well as a 2022 slides deck produced by Genuine for the NHSA available on the latter's website. Azvudine was found to inhibit some coronaviruses in vitro around 2020, leading to an interest in its use in COVID.
In his principal work, Manfred Eigen stated that the E coded by the I chain can be a specific polymerase or an enhancer (or a silencer) of a more general polymerase acting in favour of formation of the successor of nucleotide chain I. Later, he indicated that a general polymerase leads to the death of the system. Moreover, the whole cycle must be closed, so that En must catalyse I1 formation for some integer n > 1.
=== Development === The series originated from discussions between Toho producers and Yeon Sang-ho in 2018, during which Yeon selected The Human Vapor from Toho's "Transforming Human Series" for its blend of speculative science and human drama. Yeon, known for global hits like Train to Busan (2016) and Netflix's Hellbound (2021), emphasized the original film's "skillful sci-fi expressions" and emotional core, stating: "While it is a sci-fi thriller, at its core, it is a story about people. We focused on treating human emotions with care and portraying the characters’ humanity." Script development took three years, with Ryu Yong-jae—Yeon's collaborator on Parasyte: The Grey (2024)—joining as co-writer. Shinzo Katayama, who was acclaimed for his directing live action TV seriesGannibal (2022), was selected as director for his ability to merge genre elements with intimate storytelling. Katayama, upon revisiting the original, noted: "I was intrigued by its blend of human drama and romantic elements, despite featuring an absurd creature like the Human Vapor." The project represents Netflix's first partnership with Toho, aiming to revive the studio's tokusatsu legacy for global audiences using cutting-edge VFX. Wow Point, the production company behind Parasyte: The Grey, co-produces, infusing Korean expertise in genre storytelling.

Missouri and, before the abolition of capital punishment, Delaware, use or used a lethal injection machine designed by Massachusetts-based Fred A. Leuchter consisting of two components: the delivery module and the control module. The delivery module is in the execution chamber. It must be preloaded with the proper chemicals and operate with the timing of the dosage. The controller module is in the control room. This section is the portion that officially starts the procedure. The operation is done by first arming the machine and then having station members simultaneously press each of their buttons on the panel to activate the delivery. The computer then deletes the identity of the person who actually started the syringes, so the participants are not aware whether their syringe contained saline or one of the drugs necessary for execution (to assuage guilt in a manner similar to the blank cartridge in execution by firing squad). The delivery module has eight syringes. The end syringes (i.e., syringes 7 and 8) containing saline, syringes 2, 4 and 6 containing the lethal drugs for the main line and syringes 1, 3 and 5 containing the injections for the backup line. The system was used in New Jersey before the abolition of the death penalty in 2007. Illinois previously used the computer, and Missouri and Delaware use the manual injection switch on the delivery panel. Eleven states have switched, or have stated their intention to switch, to a one-drug lethal injection protocol. A one-drug method is using the single drug sodium thiopental to execute someone.
In the context of nutrition, red meat is defined as meat obtained from mammals, including beef, pork, lamb/mutton, veal, and venison. Red meat does not necessarily appear red in color. In culinary contexts, the term red meat is used more loosely to refer to meat that is red when raw. From a nutritional perspective, red meat can be a good source of protein, iron, zinc, and vitamins B1, B2, B6, and B12. However, red meat has been associated with a number of non-nutritional health impacts. According to the International Agency for Research on Cancer (IARC), unprocessed red meat probably causes cancer, particularly colorectal cancer. Studies have also linked red meat with higher risks of cardiovascular disease and type 2 diabetes. If meat is processed, such as by salting, curing, or smoking, health risks further increase. Processed meat is known to cause colorectal cancer. The World Cancer Research Fund recommends minimizing consumption of processed meat and limiting red meat to no more than three portions per week, which is equivalent to about 350–500g (12–18oz) cooked weight. Most red meat comes from farmed animals, such as cattle, sheep and pork, which are produced as part of industrial agriculture. Increased demand for red meat, especially in wealthy and emerging economies, has been a major driver of many of the negative environmental impacts of animal agriculture. Most research on the impact of food on health and environment recommends a significant reduction in meat consumption is essential to mitigate climate change and avoid the worst impacts of the biodiversity loss.
The success of the submarine is inextricably linked to the development of the torpedo, invented by Robert Whitehead in 1866. His invention (essentially the same now as it was 140 years ago), allowed the submarine make the leap from novelty to a weapon of war. Prior to the development and miniaturization of sonar sensitive enough to track a submerged submarine, attacks were exclusively restricted to ships and submarines operating near or at the surface. Targeting of unguided torpedoes was initially done by eye, but by World War II analog targeting computers began to proliferate, being able to calculate basic firing solutions. Nonetheless, multiple "straight-running" torpedoes could be required to ensure a target was hit. With at most 20 to 25 torpedoes stored on board, the number of attacks a submarine could make was limited. To increase combat endurance starting in World War I submarines also functioned as submersible gunboats, using their deck guns against unarmed targets, and diving to escape and engage enemy warships. The initial importance of these deck guns encouraged the development of the unsuccessful Submarine Cruiser such as the French Surcouf and the Royal Navy's X1 and M-class submarines. With the arrival of anti-submarine warfare (ASW) aircraft, guns became more for defense than attack. A more practical method of increasing combat endurance was the external torpedo tube, loaded only in port. The ability of submarines to approach enemy harbours covertly led to their use as minelayers.
Protactinium exists in two major oxidation states: +4 and +5, both in solids and solutions; and the +3 and +2 states, which have been observed in some solids. As the electron configuration of the neutral atom is [Rn]5f26d17s2, the +5 oxidation state corresponds to the low-energy (and thus favored) 5f0 configuration. Both +4 and +5 states easily form hydroxides in water, with the predominant ions being Pa(OH)3+, Pa(OH)2+2, Pa(OH)+3, and Pa(OH)4, all of which are colorless. Other known protactinium ions include PaOF2+, PaOF+2, PaF−6, PaF2−7, PaF3−8, PaOSO+4, PaO(SO4)−2, PaO(SO4)3−3, PaOCl2−5, and PaCl−6.

It mimics a natural hypothalamic signal that tells the pituitary to release growth hormone. The result is a rise in circulating growth hormone and, indirectly, in insulin-like growth factor 1. Over weeks of treatment this shift is associated with a selective decrease in fat stored inside the abdomen.
